Police said the trio -- aged 26, 38 and 48 -- was taken into custody after two raids in the southern state of Victoria.
The men's Names were not released by police but the 48-year-old was widely reported by Australian media to be Melbourne Cup-winning horse trainer Darren Weir.
They will be questioned as part of an investigation into alleged sporting integrity offences, including obtaining financial advantage by deception, engaging in conduct that could corrupt a betting outcome, and using corrupt information for betting purposes.
Weir, a veteran of the sport, is renowned for training the Prince of Penzance, which female jockey Michelle Payne rode to victory in the 2015 Melbourne Cup.
Racing Victoria said it had been conducting an inquiry into several people at the stables and had sought the help of police.
"Racing Victoria is committed to maintaining the highest integrity standards in our sport and, where appropriate, we will continue to work with Victoria Police on those investigations," executive general manager for integrity Jamie Stier said in a statement.
Police said a firearm and a "conducted energy device", reportedly a Taser, were also seized in the raids.
